Do the chances change if you have 40,000 people having one spin on 40,000 indentical machines or 40,000 people each taking one spin on one machine, or one person taking one spin on 40,000 machines, or one person taking 40,000 spins on one machine. I think the only factor that matters is the 40,000 spins. Am I mistaken?
The difference between fiction and reality is that fiction is supposed to make sense.

If they're all independent, I'd think it shouldn't matter.

They change only if the 40,000 people don't use the same strategy. If every play uses the same strategy, then all three cases have the same chance of getting a royal flush, as well as one you did not mention - you playing one hand on each of 40,000 different identical machines.

Correct. You have to add in "perfect strategy". If your goal is to only get a royal flush over any other hand, then the correct hold would be any card(s) contributing to a royal flush.
