These are my criteria for a room:
1. Safety and feeling safe walking to and from the hotel late at night
2. Quietness for sleeping. I will probably go to bed around 2 AM.
3. Casino with good blackjack and/or VP

Which casinos should I be looking at, how much to bet, how long to play, etc. in order to achieve my goal? I am most interested in the Nugget, but I am willing to settle for less if getting comps would be too hard.

Call the casino you wish to stay at (anywhere downtown but the Golden Nugget and you are risking a bad experience, IMHO) and ask to speak with the host on duty. Tell them you a re wiring in front money and/or ask them if you can apply for credit over the phone. They will ask you questions about your play and if you have players cards from other casinos (informing them of the level can lend some veracity to the level of play).
If you want to set something up with a host, I can put you in touch with my host at Golden Nugget. no guarantees but, if you wire money and/or get a line of credit, he will take excellent care of you.

It's ended now,(5 days ago) but recently you could get a free room and a minimum $38+ EV advantage for just getting a players card and very little play.

Blackjack: I usually play $15 and $25 min tables. My average bet would be around $30 and $50 respectively.
Craps: I play table minimum ($5 - $10) on Pass line with 2x - 20x odds
VP: 25 cent to $1
Poker: 4/8 limit or 1/3 NL

For each of these, 3 to 4 hours of play with a players card will get you sufficiently noticed for a good shot at a MIDWEEK room. I'd up things a bit for a WEEKEND room and start out with a Dealer Bet so they really notice you.
VP: 25 cent to $1
Poker: 4/8 limit or 1/3 NL
For VP and Poker.... I've no idea.
Don't expect a host to guarantee anything, they don't like to do that for anything less than black chip play.
You can use casino websites for credit applications but I think its best to do it in person if you do it at all.
Many downtown casinos have older structures wherein rooms are small and elevators noisy but things generally go well.
Las Vegas is on its own time and many tourists stay up the whole night in the casino and don't even realize it.
Four Queens has good VP and pretty fair immediate comps... Can't comment on amenities. Will personally stay there eventually.
Look at vpfree2 for vp games and the wizard's surveys for blackjack and craps. The reviews on this site are good for their pictures.
