or does it vary by Casino?

I would think more per day is always better. I know that Cesears's/Harrah's uses a term called Average Daily Theorhetical.
At my age, a "Life In Prison" sentence is not much of a deterrent.

Harrah's tiers are nationwide and based on coin-in x hours (as I'm sure you all know), with one past exception I took advantage of once; at Harrah's LV, in 1 24-hour period, I turned enough coin in a hot slot that I generated 20K coin-in while only losing $650. (got me in the end during the last hour of rating play). After I had about 12K in points, I checked with the hosts, who told me that they have a single-day exception at that property only, and if I kept playing at that rate, I would tier up. I went from Gold to Diamond in one day and it was good until March of the next year (about 9 months from when I was there). I don't know if that exception still exists, or if it's still restricted to HLV property only, but my understanding was the 1 day play tier award was significantly below the normal Diamond tier requirement. So in that case at least, I would say find out when their 24 hour clock resets (it was 3am at that time, several years ago), then make sure you keep your play within that 24 hour block and go big.
If the House lost every hand, they wouldn't deal the game.

The new tier policy is 15000 Tier for Diamond for the entire year. However, if you earn 2500 Tier in a single day, you will get a 5000 Tier credit bonus. So if you earn 2500 Tier points each day for 2 days. Then bam, you're Diamond now. I'm not sure if Diamond in a day still exists because of this new system.

The gaming day in Vegas for CET properties begins at 6am and ends at 5:59am.

As for what is better generally. You want to have the highest daily average possible, so playing big on one day is better than evenly spreading it. One possible exception to this is that you are staying at their property on a comp. Then they know you are staying at their property and will count all the days you stay as "gaming days". In that case, it may be better to spread out your gaming at least somewhat.

Let me just add something to what tringlomane said (very good, by the way):

If you happen to be playing machines at 5:59am take out your card, take a two or three minute break, and then reinsert your card or there is a danger that the computer won't recognize that the "play date" has changed. I got this tip from a TR manager at Rincon.

If you are concerned about getting the most tier points for your budget, then when you reach the bonus tier levels that meet your budget, stop playing until the next day.

The "bonus levels" are:

500 points gets a 125 bonus
1000 points gets a 1000 bonus
2500 points gets a 5000 bonus

Really good info from both of you, thanks! I have what may be a stupid question, but asking for clarification, Alan; when do the bonus points get awarded? After the day has closed, or as you cross the level goal? I'm going to try exactly what you suggest, but if the bonus levels are awarded as they are earned, I'm assuming I need to subtract them from my count mentally in order to earn a full 2500 points. Or maybe (certainly best case) if they're awarded as they're earned, and all points count, I would only need to get 1375 real points and 1125 bonus at the lower levels to make the 2500. Naw; no way they'd be that generous. I'm guessing the bonus hits, whatever level you've attained, after the day is closed in the computer. But if you'd verify that, I'd appreciate it.

I've not played anything run by computer that doesn't apply as you cross whatever point designated.

And usually, delays are openly pre-specified by the play requirements if there are any, so people aren't unpleasantly surprised when something hasn't kicked in.

(but that's not to say that's the way it is in this case, just sayin')
