How big of a sample do I need to prove something is about 1 in 6?

That largely depends on a few things. Example, if we're talking craps, where there are 36 combinations of the dice, I'd estimate that you need somewhere around 1,000 samples per combination. However, in the example of craps, there aren't really any variables to speak of, everything is of known quantity so a 7 would come up 1:6.
However, if we're talking about the lilihood of something happening, say, in a sports setting, the samples would need to be absolutely massive as there are a millllllllion times a billlllllion ways to adjust probabilities and statistics with the addition or subtraction of just one variable.
