Is their a mathematical difference between a $1 bet as compared to a $5 bet on the bonus for 21 + 3 for the house?

As long as the casino pays 9 to 1 for a win on the side bet for both wagers, the house edge as a percentage is the same.

But a bet of $5 will make you lose 5 times more in the longrun than a bet of $1, just like any other game that has a house edge.

LOL. Its probably bad enough that dealers hear that all the time but even sober players seem to say 24 and not realize what is going on.
